Test Name: FISH- X and Y Sex Chromosomes

Methodology: Fluorescence In Situ Hybridization (FISH)

Clinical Significance: To determine sex chromosome complement in individuals with ambiguous genitalia or suspected sex chromosome aneuploidy disorder. (Must be performed in conjunction with chromosome analysis.)
To determine sex chromosomes in patients who have received an opposite sex bone marrow transplant. (May be performed in conjunction with chromosome analysis but is not required.)

Testing Site: Testing will be sent to an approved reference laboratory.

Days Performed: Mon-Sat.

Turnaround Time: Routine: 3 - 5 days.


Specimen Requirements

Specimen: Blood or bone marrow

Collection Instructions: Blood or bone marrow must be received and processed by WSLH within 24 hours of collection. Specimen must be received in UWHC Core Laboratory (B4/220) on Monday-Friday only.

Collection Container: Preferred: Green top (with sodium heparin anticoagulant)

Collection Volume: Preferred: 1-2 mL blood or 0.5-1.0 mL bone marrow

Sample Analyzed: Whole Blood or Bone Marrow

Specimen Transport: Transport specimen to UWHC Core Laboratory (B4/220) at room temperature. Transport with coolant pack during hot weather (>85F).

Unacceptable Criteria: Bone marrow/blood that is clotted or hemolyzed

Stability: Ambient: 24 hours
Refrigerated: Not acceptable
Frozen: Not acceptable


Interpretation

Interpretation provided with results.

Test Limitations:

FISH does not rule out isochromosomes or other structural abnormalities of X and Y.

Additional Information:

If FISH assay is not performed in conjunction with chromosome analysis there is an additional charge for processing the specimen.
