Methodology: Microscopy

Days Performed: Daily, 0700 - 2230.

Turnaround Time: Routine: Smear/Stain: Calcofluor white stain 8 hours. Hair, skin and nails may take up to 24 hours.

Specimen Requirements

Specimen: Skin scrapings from periphery of lesion, nails, hair, sputum, tissue, urine, esophageal brush or body fluids

Patient Preparation: Standard aseptic collection procedures should be followed.

Collection Instructions: Rinse mouth thoroughly before collecting sputum. Clean skin with 70% alcohol for skin scrapings. Sputum, tissue, or 2.0 mL of body fluids in sterile, screw top container. Urine is appropriate only when disseminated mould (not Candida) infection is suspected. Skin scrapings from periphery of lesion, nails or hair should be submitted in a sterile screw top container or between 2 glass slides.

Collection Container: Preferred: Sterile screw cap container

Specimen Transport: Transport specimen to UWHC Microbiology (B4/231).

Stability: Ambient: Hair/skin/nails- 3 days; Other- 2 hours
Refrigerated: Hair/skin/nails- Not acceptable; Other- 7 days
Frozen: Not acceptable


Expected Results:

No Fungi found in direct microscopic examination.
